Cross-GPCR Database Statistics

Database-level summaries across 283 Class A GPCRs: receptor rankings, TM-domain patterns, recurrent generic-numbered positions, and variant annotations.

A. Cross-GPCR Comparison

B. Transmembrane Domain Patterns

C. Core Functional Residues (CFRs)

Positions that repeatedly show above-threshold ΔRRCS changes across many GPCRs, suggesting recurrent activation-associated contact remodeling.

What are Core Functional Residues (CFRs)?

Residues that carry a GPCRdb generic number are mapped onto that shared position across the 283 receptors. A position is flagged as a CFR if it appears in the above-threshold contact list of many receptors, meaning that position repeatedly participates in active-inactive contact remodeling across the family. The mapping used here comes from the full GPCRdb residue maps for all 283 receptors, the same maps used for the manuscript analysis.

Positions are ranked by a composite score of (1) how many receptors show an above-threshold change at that position (recurrence) and (2) the average magnitude of change (mean |ΔRRCS|). High-ranking positions mark generic-numbered sites that recur across receptors and may reflect conserved components of Class A GPCR activation-associated contact networks.

The ranked list below is cut at the top 50 positions, the cutoff used in the manuscript analysis. That cutoff is a reporting threshold, not a definition: 368 positions meet the CFR criterion of recurring in at least three receptors. A position ranked below 50 is still a recurrent CFR position, it just falls outside the subset reported here.

Where these cross-receptor numbers come from. Cross-receptor recurrence analyses are limited to residues assigned GPCRdb generic numbers. Residues without one, largely in loops and terminal regions, cannot be aligned to a shared position across receptors and take no part in these statistics. Recurrence statistics on this page are the results submitted with the manuscript, computed on the full GPCRdb residue maps for all 283 receptors. The per-receptor annotation used to build individual report pages numbers only residues that took part in a high-magnitude contact, so it yields lower recurrence counts. Individual receptor reports are unchanged and keep their own report-generation data.

D. Variant Correlation

Variant analyses include 279 receptors. NPY6R and TAAR3 lack protein-coding missense annotations because they are annotated as pseudogene loci. GP182 and P2RY8 are excluded from the current variant dataset because their identifiers were not successfully resolved during data collection.

AlphaMissense pathogenicity at the top 50 recurrent positions

Among decisive AlphaMissense predictions, 67.0% were classified as pathogenic at the 50 most recurrent generic positions, compared with 44.1% at other generic-numbered positions: odds ratio 2.58 (95% CI 2.42 to 2.76). Adjusting for conservation reduces the odds ratio to 1.52 (95% CI 1.41 to 1.65).

Ambiguous predictions are shown in the chart but excluded from the odds ratio. 50 most recurrent positions: n = 7,445. Other generic-numbered positions: n = 11,150. The comparison group is every other generic-numbered position, which includes recurrent positions ranked below the cutoff.

This association supports recurrence as a variant-prioritisation feature. It does not establish pathogenicity.
